Regulatory T cells

Studies from the Maizels Lab and others have established the prominent role of regulatory T cells (Tregs) in facilitating and maintaining helminth infections. Our most recent review by McManus and Maizels (2023) is here.

Key advances in the field from our lab have been:

In vivo role of Tregs:

Tregs expand in H. polygyrus infection in mice (Finney et al, 2008), and depletion of Tregs renders mice resistant (Smith et al., 2016). In filarial nematode infection, Brugia malayi also expands TYregs (McSorley et al 2008), while in infection with Litomosoides sigmodontis, combination treatment with anti-CD25 with anti-CTLA-4 or anti-GITR elicits parasite killing (Taylor et al 2005, 2007).  Tregs also mediate bystander suppression of allergy in mice (Wilson et al 2005).

Treg expansion driven by parasite products: H. polygyrus ES induces the canonical Treg transcription factor Foxp3 in naive T cells, phenocopying TGF-β (Grainger et al 2010).

Tregs are now understood to play an integral role in most parasitic diseases, but can act to facilitate infection and/or protect against pathological outcomes, depending on the host/parasite combination (McManus and Maizels 2023)
